Overview
CJC-1295 with DAC is a modified GHRH(1-29) analogue engineered with a lysine substitution and a maleimidoproprionic acid (MPA) linker that forms a covalent bond with circulating albumin. This albumin binding dramatically extends its half-life, allowing for infrequent dosing intervals in research protocols. The extended action produces a sustained "bleed" of GH elevation rather than a sharp pulse.
CJC-1295 without DAC (also called Mod GRF 1-29) is a distinct compound with a shorter half-life (~30 minutes) that produces more physiological GH pulses when timed appropriately. Both variants are widely used in research. CJC-1295 with DAC is notable for studies examining the effect of sustained GH elevation on body composition, recovery, and aging biomarkers.
Mechanism of Action
CJC-1295 stimulates GHRH receptors on pituitary somatotrophs. The DAC modification allows covalent binding to albumin's Cys34 residue, creating a circulating depot that slowly releases the active peptide. This results in sustained elevation of GH and subsequently IGF-1 with a once-weekly or twice-weekly dosing profile in research settings.
