Overview
AOD-9604 was developed by Metabolic Pharmaceuticals as a lipolytic fragment of hGH. The fragment spanning residues 176β191 of hGH had been identified as the region responsible for the hormone's fat-burning activity. By isolating and modifying this region, researchers aimed to capture the metabolic benefits while avoiding the insulin resistance and IGF-1 elevation associated with full hGH use.
In animal studies, AOD-9604 reduced body fat, stimulated lipolysis, and inhibited lipogenesis without affecting blood glucose or IGF-1. A modified version (with an additional tyrosine residue to enhance stability) was taken into human clinical trials, though these trials showed mixed results in obese subjects compared to the robust animal data. It is now a subject of ongoing investigation in cartilage repair and metabolic research.
Mechanism of Action
AOD-9604 acts on beta-3 adrenergic receptors in adipose tissue to stimulate fat breakdown. It also appears to inhibit lipogenesis through mechanisms distinct from full hGH. Critically, it does not bind to the GH receptor with sufficient affinity to stimulate IGF-1 production or promote glucose intolerance, making its metabolic profile distinct from intact hGH.
